嗜碱性短小芽孢杆菌碱性蛋白酶的研究——Ⅲ.酶的性质及应用

【摘要】 本文对嗜碱性短小芽孢杆菌(Alkaliphilic Bacillus pumilus)B45菌株产生的碱性蛋白酶性质进行了研究。该酶对酪蛋白水解的最适 pH 为10.0—10.5,在 pH7—10之间稳定,最适反应温度为45℃,稳定性较好。最适底物浓度为1%。四硼酸钠、氯化钙对酶有激活作用,三聚磷酸钠及碳酸钠在常温下对酶活力无影响,而在40℃时对活力略有抑制,加入0.2g/LCaCl2可以保护酶的活性。B45蛋白酶对血,奶污布的去污效果明显,其去污力比一般洗衣粉高10—13倍。

【Abstract】 This paper has examined some propertiesof alkaline proteinase from alkaliphilic Ba-cillus pumilus B45 strain.The optimum pHand temperature for the casein hydrolysiswere 10.0—10.5,and 45℃,respectively.Theenzyme was stable at pH ranged from 7—10,and at temperature under 45℃ for 2h.About 80% of its activity was remained at50—55℃ for 30min.and 40% at 60℃ for30 min.The concentration of 1% casein wasthe best on the proteolytic effect.The sodiumtetraborate,calcium chloride enhanced acti-vity of the enzyme,but sodium tripolyphos-phate and sodium carbonate inactivated en-zyme at 40℃,Ca2+of 0.2g/L protected en-zyme against inactivation.Apparently,addi-tion of enzyme into detergent was good forwashing dirt of blood and milk. The ef-ficiency of detergent was 10—13 times higherthan the detergent without enzyme.
