分泌抗-HBe和抗-HBc单克隆抗体杂交瘤细胞系的建立和鉴定
Establishment and Identification of Monoclonal Antibody Hybridoma Cell Lines of Anti-HBe and Anti-HBc
【摘要】 应用具有 HBeAg 和 HBcAg 双抗原活性的基因工程菌表达产物的粗提物免疫 BALB/c小鼠后,取脾细胞与小鼠骨髓瘤细胞系 SP2/0融合,分别筛选出4株抗 HBe 单克隆抗体杂交瘤细胞株(WHBe1,WHBe2,WHBe3,WHBe4)和4株抗 HBc 单克隆抗体杂交瘤细胞株( WH-Bc1,WHBc2,WHBC3,WHBc4)。经3个月连续培养,能稳定分泌效价高、特异性强的单克隆抗体。
【Abstract】 We successfully established 4 hybridoma cell lines of anti-HBe and 4hybridoma cell lines of anti-HBc,and the specificity and titer of the monoclonal anti-bodies secreted by the cell lines were not influenced after cultured for 3 months con-tinually.Specificity test showed that the monoclonal antibody has very high speciality of anti-HBe and anti-HBc.The class and subclass of Ig and chromasome of hybridoma cells werealso identified.
