【摘要】 本文建立了快速简便测定人体血浆中氯唑沙宗的反相高效液相色谱(RP-HPLC)分析法。采用YWG-C18H37柱,甲醇-水(60:40,V/V)为流动相,内标为非那西丁,UV检测波长282nm。血浆样品用乙酸乙酯提取,色谱蜂分离完全,方法线性良好,r=0.9999,提取回收率为94.44±:2.02%(n=20),方法回收率为103.18=2.68(n=20)。天内精密度以CV表示为1.94%~3.38%,天间精密度为2.91%~4.19%。本法用于测定人体血浆中氯唑沙宗的浓度。

【Abstract】 An EP-HPLC method for determining plasma levels of chlorzoxazone (5-ehloro-2(3H)-benzoxazolone), a potent skeletal muscle relaxant, effective in the treatment of skeletal spasms, is presented. A reversed phase column packed with YWG C18 (10μm, 160×5mm, id.)with isocratie mobile phase of 60% methanol in water was used. The eluent was monitored with an UV detector at 282 nm. Phenacetin was chosen as the internal standard.The calibration curve in the range of 0.507-40.56μg/ml showed good linearity (r = 0.999 9), and the detection limit using 1ml plasma was 30 ng/ml. The HPLC method utilized a rapid and inexpensive extraction procedure. Using ethylacetate, the extraction efficacy of chlorzoxazone from plasma was 94.44 ±2.03%. The recovery of chlorzoxzaone was 103.18±2.68%, Coefficients of intra-day and inter-day variations were less than 3.37% and 4.19% respectively.The method was used to profile the plasma levels of chlorzoxazone.
