免疫印迹法检测抗核糖体抗体及其临床意义
DETECTION OF ANTIBODY TO RIBOSOMES BYIMMUNOBLOTTING TECHNIQUE AND ITSCLINICAL SIGNIFICANCE
【摘要】 抗核糖体抗体主要见于 SLE,对 SLE的诊断具有一定特异性。本文用免疫印迹法(IBT)证实兔胸腺 ENA 中38,16.5,15KD 多肽具有核糖体抗原表位,应用兔胸腺ENA代替胞浆纯化核糖体用 IBT检测抗核糖体抗体,结果表明该抗体在40例 SLB中的阳性率为 15%,在抗Sm和/或抗 RNP抗体阳性的 SLE患者中,阳性率可达 30.8%,而在 143例非 SLE风湿病患者中阳性率仅1.4%,差别显著(p<0.01)。本文SLE中抗核糖体阳性与阴性二组在主要临床特征上无显著差别(P>0.05)。
【Abstract】 Antibody to ribosomes is present in patients with SLE and relatively specific for the diagnosis of SLE. In this paper, we used saline extracts of rabbit thymus acetone powder instead of purified ribosomes from cytoplasma as antigen and detected the antibody to ribosomes in patients’ sera with IBT. We found that antibody to ribosomes identified three peptides with molecular weight 38KD, 16.5KD and 15KD respectively. The positive rates of anti-ribosome were 15% and 1.4% (p<0.01) in patients with SLE (n = 40) and those without SLE (n = 143) respectively. In SLE patients with positive anti-Sm and/or anti-RNP, the positive rate of anti-ribosome could even reach 30.8%. But in this paper we couldn’t draw any associations between antibody to ribosomes and any particular clinical feature of SLE.
