内蒙古地区鸡新城疫病毒分离株特性的研究

【摘要】 在内蒙古自治区五个盟市十个养鸡埸中从发病、死亡鸡中分离出8株鸡新城疫病毒,並定名为HH1、WH、HH2、HH3-、HH4、DS、JN和HH5。对此8株NDV分离物电镜下的形态、毒力、蚀斑特性、血凝素的热稳定性、对部分动物红细胞的凝集能力、凝集解脱特征及对福尔马林的敏感性等生物学特性进行了系统的测定;并用新城疫单克隆抗体对NDV分离物进行了初步的抗原性分析。于此同时,并与攻毒株(F48E8、NM株)和疫苗株(Ⅰ系、Ⅱ系和LaSota株)作了比较研究。分离的8株NDV致病性测定结果表明:MDT为42.0~60.2小时;ICPI为1.61~1.98;INPI为1.02~2.43,全部属于速发型NDV。8株NDV均能在鸡胚成纤维细胞(CEF)上产生蚀斑。本试验发现了两株对热相当稳定的毒株。各NDV分离株内部以及与现行ND疫苗株和NDV参考株之间存在着一定程度的抗原性差异。

【Abstract】 The morphology, virulence, plaque property, some other biological properties, antigenicity of eight Newcastle Disease Virus isolated from Inner Mongolia (HH1,WH,HH2,HH3,HH4, DS JN & HH5) and control strains were investigated systematically. By the results of MDT/MLD(42.0~60.2hr),ICpI(1.61~1.98),IVPI (1.02~2.43)and plaque formation on CEFS, It was found that eight isolateas all fall into Velogenie NDV Strains. The biological properties of strains tested by thermostability of hemagglutinin, agglutination of certain mammalian erythrocytes agg- lutination elution pattern and susceptibility to formalin were, to a certain extent, variable among different strains testes. Use of Monoclonal antibodies in the Indirect Immunofluorescence assay(ⅡF)and hemagglutination inhibition(HI)tests showed antigenic variation among strains tested. Suming up the results of X2~test, in ⅡF test and variance analysis in HI—test, the WH, HH3 and HH4 strains would be important for the selection of seed strain for inactivated vaccine.
