光生物素DNA探针的制备及其检测恶性疟原虫DNA的研究
PREPARAlION UF PHUTOBIOTIN-DNA PROBES AND IHEIR USE FOR DEIECTION OF PLASMODIUM FALCIPARUM DNA
【摘要】 本研究使用光生物素(PHOTOPROBETM Biotin)标记恶性疟原虫(P.f.)全基因组DNA,含P.f.cDNA片段的重组质粒DNA(C7)及克隆的P.f.cDNA片段(P9-25)等作为探针,并以斑点杂交试验检测靶DNA。实验结果显示,光生物素标记法简单、快速和安全;标记的探针活性较高,于-20℃贮存10个月无明显降低。用生物素化的P.f.全基因组DNA及C7探针分别可检出低至125pg和250pg的纯化的P.f.DNA。
【Abstract】 In order to develop a simple,more rapid and cheaper system for diagnosis of malaria in field and clinic,parasite genomic DNA isolated from cultured Plasmodium falciparum(P.f.), recombinant plasmid DNA containing P.f.cDNA fragments(C7)and cloned P.f.cDNA fragments(P9-25)were labeled with PHOTOPROBE TM Biotin,and some of them were used as probes to detect target DNA by dot blot hybridization.The results obtained from the studies show that the photobiotin labeling method is simple,rapid and safe.The labeled probes showed high activity that did not reduce apparently when they were stored for I0 months at -20℃.The biotinylated P.f.genomic DNA and C7 probes could detect 125 and 250 pg of purified P.f. DNA,respectively.
