玉米醇溶蛋白基因Z15转化石刁柏的研究
STUDIES ON THE TRANSFORMATION OF THE Z15 GENE OF ZEIN TO ASPARAGUS
【摘要】 利用含有玉米醇溶蛋白基因(Z15)的重组Ti质粒感染石刁柏茎段,诱导其产生愈伤组织,并使愈伤组织在MS+0.5 mg/1 NAA+1.5 mg/1 KT的分化培养基上产生不定芽和再生植株,不定芽的诱导率提高了76.7%。经纸电泳测定和卡那霉素抗性鉴定证明,石刁柏的再生植株能够合成胭脂碱,并能在附加200 mg/1卡那霉素的上述培养基上正常生长。
【Abstract】 The stem segments of Asparagus officinalis L. were infected by Ti plasmid (of Agrobacterium tumefaciens) reconstituting with the Z15 gene of zein and were induced to produce callus. The adventitious buds and regenerated plants were induced from the callus on the diffe-rentiation medium of MS+0.5mg/l NAA+1.5mg/l KT. The rate of induced adventitious buds was increased by 76.7%. The results of paper electrophoresis analysis and Kanamycin resistance test showed that the regenerated plants of Asparagus could synthesize nopaline and grow normally on the medium mentioned above added with 200 mg/l Kanamycin.
