应用捕捉法ELISA检测乙肝患者血清中IgG类HBsAg特异性免疫复合物

【摘要】 本文建立了一种检测HBsAg/IgC—CIC(Circulating ImmuneComplex)捕捉法ELISA(Enzyme Linked Immunosorbert Assays)。该法特异性强,敏感性高,重复性好,避免了固相抗HBs双抗体ELISA受血清中游离HBsAg的抑制作用而影响HBsAg/IgG—CIC检出的缺点。并行检测HBsAg/IgG—CIC和血清C3发现,乙肝患者血清中HBsAg/IgG—CIC检出率血清C3下降显著高于非下降组,提示HBsAg/IgG—CIC可能是导致乙肝患者血清C3下降的又一重要因素。

【Abstract】 An assay for the specific detection of HBsAg/IgG-CIC by ACCA-ELISA (antibody class capture assay-ELISA) was reported in this paper. This method is not only simple, specific, sensitive and reproducible, but also free of the weakness of inhibiting detection of HBsAg/IgG-CIC by serum HBsAg which existed in the ELISA using anti-HBs attached on solid phase. The result that both C3 and HBsAg/IgG-CIC were detected in one serum sample indicates that positive rates of specific HBsAg/IgG-CIC in the patients with low level of serum C3 were signifcantly higher than in those with normal level. It suggests that HBsAg/IgG-CIC be one of important substances that make serum C3 reduced decline.
