固定化增殖酵母工业应用的研究——Ⅱ、分批和连续发酵生产乙醇试验研究

【摘要】 本文对固定化增殖酵母分批和连续发酵生产乙醇进行了试验研究。试验结果表明,在分批发酵中,PVA固定化增殖酵母的乙醇生产能力是游离酵母的3倍,达到24毫克/毫升凝胶/小时以上。发酵周期比游离酵母缩短了一半。在连续发酵中,发酵培养基在柱中停留时间3小时,发酵流出液中的乙醇含量平均在7.1%(v/v)以上,固定化增殖酵母的乙醇生产能力平均达到了18毫克/毫升凝胶/小时以上,是传统发酵方法的8~10倍。同时,通过分批发酵22批次(重复使用)和连续发酵120天的稳定性试验证实,PVA固定化增殖酵母的活性和机械强度能长期保持不变,表现出良好的反应稳定性,具有较高的工业应用价值。

【Abstract】 This paper deals with the experiment on alcohol production by immobilized multiplying yeast cell in batch and continuons fermentation systems. The experimental results showed that in the batch fermentation a high productivity (24 mg/ml gel/hr) was achieved by using immobilized multiplying yeast cells entrapped in PVA gel,which was 3 times as high as that using free yeast, and the fermentation time was half shortened; in continuous fermentation, the alconol productivity was at the level of 18mg/ml gel/hr, which is 8-10 times as high as that in traditional fermentatiom process,and the fermentation liquor could be maintained in the columns for 3 hours and the average alcohol concentration in it was found to be above 7.1%, It was also fornd that after 22 batch fermentation (repeated using the yeast cells entrapped in PVA gel) and 120 days continuons fermentation respectively, the viability and mechanical strength of the immobilized multiplying yeast cells entrapped in PVA gel was well retained and had good reaction stability for industrial application.
