国产甲氟哌酸体外抗菌作用的研究
IN VITRO ACTIVITY OF PEFLOXACIN COMPARED TO OTHER ANTIBIOTICS
【摘要】 甲氟哌酸(Pefloxacin)对需氧的革兰氏阴性、阳性细菌显示了广谱的抗菌效应。与九种抗菌药体外抗菌活性比较显示:对201株金黄色葡萄球菌的MIC50、MIC90分别为0.25mg/L、1.0mg/L。对多数肠道革蓝氏阴性杆菌(包括大肠杆菌、沙门氏菌属、志贺氏菌属其MIC90为0.25mg/L,对变形杆菌属为2mg/L。对201株绿脓杆菌的MIC50、90分别为2mg/L、8mg/L。其抗绿脓杆菌作用与氧哌嗪青霉素相似,而更优于头孢氨噻肟与庆大霉素。 体外杀菌曲线也显示了该药在4倍的MIC浓度下在8小时及12小时可全部杀灭绿脓杆菌、大肠杆菌及痢疾杆菌。
【Abstract】 The in-vitro activity of pefloxacin was compared with that of two other quinolones, five beta-lactam antibiotics, tetracycline and gentamicin against 1244 clinical-isolated strains. The results showedthat the clinical pathogens tested were highly susceptible to pefloxacin. The MIC50 and MIC90 of pe-floxacin and norfloxacin for each strain were the same, but greater than that of pipemidic acid (8-64fold). Pefloxacin was highly active against 205 strains of Staghylococcus aureus (MIC50 0.25 mg/l,MIC90 1.0 mg/l). The MIC90 of Pefloxacin against 791 strains of Enterobacteriaceae ranged from 0.25mg/l (E. coli, Klebsiella spp, Salmonella spp and Shigella spp.) to 2 mg/l (Proteus spp). In vitrostudy, the susceptibility tests of 201 strains of Pseudomonas aeruginosa showed that the MIC50 andMIC90 of pefloxacin (2 and 8 mg/l) were similar to piperacillin (4 and 16 mg/l), but more activethan that of cefotaxime (16 and 64 mg/L) and gentamicin (8 and>256 mg/l). Pefloxacin was superiorto cefazolin, cefotaxime, ampicillin, piperacillin, tetracycline and gentamicin in the investigation againstStaph aureus. No enterobacteria were resistant to pefloxacin, while some of them were resistant tobetalactams and gentamicin. Time-killing curves showed that pefloxacin at concentrations four-time bigger than its MIC decre-ased the viable counts by 3 logs or more within 18 hours. A 2 to 16-fold increase or MIC wasobserved by increasing the inoculum 1000-fold with most of the strains tested.
