草鱼出血病病毒武汉南湖株的精细结构与基因组及多肽的研究
FINE STRUCTURE, GENOME AND POLY-PEPTIDES OF A GRASS CARP HEMORRHAGE VIRUS ( GCHV ) ISOLATE FROM THE SOUTH LAKE IN WUHAN HEMORRHAGE
【摘要】 对草鱼出血病病毒(Grass carp hemorrhage virus,GCHV)武汉南湖株的精细结构的研究表明,该病毒具廿面体对称结构,由双层衣壳组成,直径为72nm。外层衣壳上的子粒为中空型,分为五邻体和六邻体。五邻体直径11nm,中心孔径6nm。六邻体直径12.6nm,中心孔径7.5nm。内层衣壳直径50nm,其上附着有长7.5nm、内径6.4nm、外径10.7nm的圆柱形钉状物.病毒基因组为dsRNA分段基因组,由11个片段组成,分为4组,分子量分别是:2.75×10~?d、2.40×10~?d、2.34×10~?d、1.35×10~?d、1.32×10~?d、1.26×10~?d、0.93×10~?d、0.81×10~?d、0.71×10~?d、0.58×10~?d、0.56×10~?d。病毒多肽组份有11个。分别是VP130(VP表示病毒多肽。130为分子量,即130×10~3d,下同)、VP120、VP113、VP107、VP75、VP65、VP60、VP52、VP42,VP39和VP31,其中VP130、VP120、VP60、VP39和VP31为多肽的主要组份。病毒的内层衣壳具有6个多肽组份,分别是VP130、VP120、VP113、VP107、VP75、和VP39,主要组份是VP130、VP120和VP39.外层衣壳所特有的多肽是VP65、VP60、VP52、VP42和VP31.我们认为草鱼出血病病毒武汉南湖株是呼肠孤病毒科的一员。
【Abstract】 GCHV ( grass carp hemorrhage virus) was isolated from the South Lake in Wuhan, Hubei. Studies of the fine structure reveal icosahedral viral particles approximately 72 nm in diameter and composed of a double capsid. The viral cores, lacking the outer shell capsoraeres, are about 50. nm in diameter and show regularly spaced projections. Identification of viral nucleic acid has shown that it is dsRNA. By SDS-PAGE it has been determined that the viral genome consists of 11 segments of RNA di-vided into four size classes. I. l.(Mw 2.75×106D), 2.(Mw 2.40×106D), 3.(Mw 2.34×106D); II. 4. ( Mw 1.35×106D), 5. (Mw 1.32 × 106D), 6.(Mw 1.26×106d), II. 7. (Mw 0.93× 106d ), 8. ( MM- 0. 81 ×106d ) , 9. (Mw0.71 ×106D), IV. 10. (Mw 0.58×106D), 11. (Mw 0.56×106D). The capsid polypeptides of the intact virions and viral cores were analyzed by SDS-PAGE. The intact virions contain 11 species of polypeptides, which are divided into three groups. They are I. VP130, VP120, VP113, VP107;II, VP75, VP65, VP60, VP52, III. VP42, VP39, VP31. Among them VP130, VP120, VP60, VP39, VP31 are the main components. The viral cores have 6 species of polypeptidas, they are VP130, VP120, VP113, VP107, VP75, VP39 and the main components are VP130, VP120, VP39.From the fine structure of the virion and the composition of the viral genome and polypeptides, we come to the conclution that GCHV isolate from the South Lake in Wuhan is a member in Reoviridae.
