【摘要】 采用修改的¹⁴C-GABA法测定小鼠脑、肝、肾组织GABA-T活性,方法灵敏可靠。氨基半乳糖诱发的肝性脑病小鼠肝脏GABA-T活性明显降低。首次报道了小鼠肝匀浆上清液对脑GABA-T有抑制作用,经加热、盐析、Sephadex G-150层析,抑制活性在洗脱前段。提示严重肝损害时肝脏对GABA的分解代谢降低,正常肝细胞中存在有对脑GABA-T有抑制作用的分子量较大的物质更多还原

【Abstract】 The GABA-T activities of mice brain, liver and kidney were determined by the modified labeled "C-GABA method, and the method has been proved sensitive and feasible.Hepatic encephalopathy in galactosamine-induced mice was associated with a marked decrease of GABA-T activity of liver. The inhibition of brain GABA-T activity by the LHS of normal mice was demonstrated for the first time.After heat denaturation at 55℃ and ammonium sulfate fractionation, the elution pattern of Sephadex G-150 gel- filtration showed the inhibitory activity localized just behind the exclusive volume. These findings indicate a decrease of GABA catabolism in liver in severe liver injury, and there is a kind of GABA-T inhibitor in LHS with rather big molecular weight更多还原
