利福喷丁和苯巴比妥诱导小鼠肝细胞滑面内质网增生的作用

【摘要】 利福喷丁是新型长效抗结核抗生素,通过透射电镜观察,发现它和典型的肝微粒体酶诱导剂苯巴比妥均能显著诱导小鼠肝细胞滑面内质网的增生。由于这种作用常与肝微粒体酶诱导作用有关,故本文从形态学方面证实了利福喷丁和苯巴比妥对肝微粒体酶具有诱导作用。

【Abstract】 Rifapentine(R773, DL473) is a new long-acting antituberculousdrug and phenobarbital(PB) is a well known potent inducer of hepatic mixed function oxidase system. Male mice( 5 in each group) were given orally R773 20, 40, 80, 120 mg/kg or PB 30 mg/kg, respectively, once a day for 15 days. Then, observed the structure of the liver by the transmission electron microscopy, significant proliferation of mice hepatocyte smooth endoplasmic reticulum (SER) in PB group and a dose-dependent proliferation of SER in R773 groups were found following pretreatment with these two drugs. In treated groups, SER usually appeared as circular or oval vesicles and often showed a considerable degree of dilatation. Beside these, significant increase in amount of SER and the area occupied by SER in cytoplasm were also observed. The proliferation of SER induced by drugs is usually related to the inductive effects of drugs on hepatic mixed function oxidase system, so it has been supported that R773, like PB, is a potent inducer of hepatic drug metabolizing enzymes in mice.
