抗胃癌单克隆抗体与不同血卟啉光敏剂交联物的制备与鉴定
Preparation and identification of monoclonal antibody against gastric cancer and hematoporphyrin photosensitizer conjugates
【摘要】 在交联剂碳化亚胺作用下,把血卟啉(HP)、血卟啉衍生物(HPD)或双血卟啉醚(DHE)与抗胃癌单克隆抗体3H11交联,经薄层层析、紫外光谱、红外光谱和高效液相色谱鉴定,证实已形成3H11-HP、3H11-HPD 和3H11-DHE 三种交联物。HPD 和 DHE 与3H11比 HP 更容易交联。在三种交联物中以3H11-HPD 对 BGC-823细胞的体外杀伤作用最强。
【Abstract】 Hematoporphyrin(HP),Hematoporphyrin deriv-ative(HPD)or Dihematoporphyrin ether(DHE)wereconjugated to monoclonal antibody 3H11 against g-astric cancer through 1-ethyl-3-(3-dimethyrlam-inopropyl)carbodimide.in thin (?)ayer chromatogr-aphy,ultraviolet spectrophotometry,infrard sp-ectrophotometry and high pressure lipid chro-matography studies showed that three conjugates3H11-HP,3H11-HPD and 3H11-DHE had been f-ormed the conjugation of HPD and DHE with3H11 was easier than that of HP.The 3H11-HPD caused a greatest killing effect onBGC 823 cell line in vitro in three conjugates.
