肾Na·K-ATP酶在热环境中的变化及其调节作用
CHANGES OF RENAL Na·K-ATPase AND ITS REGULATION IN THE HEAT
【摘要】 本研究观察了热应激和热习服大白鼠肾Na·K-ATP酶的变化及不同热应激时间和应激后不同时间对大鼠肾Na·K-ATP酶的影响,并进行了醛固酮对该酶调节作用的离体实验。结果表明:大白鼠热应激时肾Na·K-ATP酶没有明显变化,应激后24h测定值较对照升高1.3倍。习服大白鼠热暴露时,酶活力显著升高,并随习服期延长,变化幅度渐小且趋于稳定。延长热应激时间,24h后酶活力测定值随醛固酮浓度进一步升高。离体实验中,在醛固酮对酶最大作用浓度4×10-10M下,1.5h后,见酶活性显著升高。结果提示:热环境中,肾Na·K-ATP酶受血浆醛固酮浓度调节而规律性地变化,并参与对尿液Na+、K+含量的调控。
【Abstract】 Adult male Wistar rats were used in this study to observe the change of renal Na· K-ATPase and its regulatory role in the heat. The effect of aldosterone on renal Na· K-ATPase in vitro was also observed. The results indicated that renal Na·K-ATPase activity of rats was not changed immediately during heat stress, but did increase 1.3 time when observed 24 hr. later. In the heat acclimation period, the increase of renal Na· K-ATPase was observed immediately after heat exposure, and it gradualy diminished and reached to a steady state. The increments of aldosterone and Na·K-ATPase activity were greater if the time of heat exposure prolonged. In vitro, the optimal concentration of aldosterone for a maximal Na·K-ATPase activity was 4×10-10M, and the increase of Na·K-ATPase activity occurred until 1.5h after incubation. The results suggest that in heat renal Na·K-ATPase, its activity is regulated by aldosterone and changes regularly, participates in regulation of sodium and potassium of urine.
