硒缺乏实验性心肌病的电显细胞化学研究

Ultrastructural studies on pathogenesis in nutritional cardiomyopathy of selenium deficient rat with electron microscopic cytochemistry

【摘要】 本文用低硒半合成饲料和补硒合成饲料喂养大鼠,喂养8和12周后分别处死。对心肌标本作常规电镜和细胞化学(细胞色素氧化酶、酸性磷酸酶和 Ca++ATP 酶)检查。实验结果表明硒缺乏可引起心肌下述主要改变,心肌膜系呈现不同程度损伤;心肌氧化磷酸化障碍,氧利用降低,ATP 合成减少;某些细胞器内 ATP 依存性钙的调控运转异常,从而进一步导致细胞器和收缩成分的一系列改变。

【Abstract】 Twenty four male rats of three weeks after weanling(44—52gm)were used inthis study.Twelve rats of them were fed on selenium-deficient diet(less than 0.05ppm)composed of Torula yeast,sucrose and vitamins as an experimental group.Another twelve rats were fed on supplementary diet with 0.25Ppm selenium.Aftertwo and three months,experimental and control rats were sacrificed respectivelyand the hearts were removed immediately.The tissues obtained from left ventricularfree,wall were examined by conventional and cytochemical electron microscopy.The growth of the rats in experimental group was inhibited by selenium deficie-nt diet and significant difference(P<0.05)was noted between control and expe-rimental group after nine weeks.However,amount of feeding was not different ineach groups even after nine weeks.Swelling of mitochondria,edematous and degenerated myocardial cells,dilatation ofsarcoplasmic reticulum were recognized in the observation of conventional electronmicroscopy.Disarray of myofibril and increasing mitochondria were observed afterthree months of feeding.Activity of cytochemical reaction was decreased in mitochondria of experimentalgroup in cytochrome oxidase and Ca++-ATPase.Experimental group showed increa-sing tendency of lysosome contained acid phosphatase in count,but the activity,but the activityof acid phosphatase was decreased.Ruthenium red positive material in experime ntalgroup was scattered on the sarcolemma,suggesting that distribution of the muco-polysaccharde was unevenly dispersed.From these finding it is strongly suggestedthat cytochrome oxidase acid,phosphatase and Ca++-ATPase changes are closelyrelated to the necrotic process of myocardial cell in selenium deficient rats.
