光敏感核不育水稻花粉败育过程中花药的差示扫描量热法(DSC)研究

【摘要】 本文以湖北光敏核不育水稻(Hubei Photoperiod Sensitive Genio Male—Sterile Rice i·e·HPGMR)农垦58A和常规品种农垦58为材料,在人工控制的长,短日照条件下,用DuPont9900型差示扫描量热计测取了不同发育时期花药的DSC谱。结果表明:可育花药的DSC谱上的低温吸热峰面积随发育呈现周期性变化;不育花药在单核中期以前与可育花药差别不大,而在单核中期以后的DSC谱上的低温吸热峰面积明显小于可育花药,且不呈现周期性变化,而逐渐减小到接近对零。

【Abstract】 Hubei photoperiod Sensitive Genie Male-Steril Rice (i. e. HPGMR) Nongken 58A was discovered in 1973. This material is characterized by the basic spaciality to be sterile under long-day sunshine and to be fertile unber short-day sunshine, which is discovered for the first time at home and abroad and is of great significace in its application and theoritical researches.This experiment adopts HPGMR 58A and normal variety of Nongken 58, in which DSC curves in different developing stages of anthers of 58ALD, 58LD and 58SD (LD relers to long-bay sunshine, SD rkfers to short-bay sunshine)are measured and obtained by means of Dupont 9900 DSCFig. 1 shows DSC curves of anthers, each showing a series ofendotncrmic peaks at 45--90C which may be due to the denatura-iiou of DNA,RNA, proteins, starches and many other substances within the anthers. The difference in curves reflects the content of these substances.Table 1 and Fig. 2 show: (1) There is--little difference ine ndoth-ermic amount between anther of 58ALD before one-celled pollen middle stage and anthers of 58ASD, 58LD and 58SD, which shows 58ALD,can perform normal meiotic division of the pollen mother cell; (2) the endothermic amount of anther of 58ALD is evidently lower than those of 58ASD, 58LD and 58SD, and decreases close to zero in straight line. This reveals that the pollen abortion of HPGMR 58A starts in the one-celled pollon middle stage;(3)there appears peak value in the endothermic amount of anthers of 58ASD, 58LD and 58SD in the meiotic division stage of pollen mother, the one-celled pollen early stage and two-celled pollen late stage, which shows there is a large amount of substances, such as DNA, RNA,proteins and so on, within the pollen in each stage, and it lays basis of substance for the meiotic division of the pollen mother cell ooe-celled division and two-celled division. The above conclusion basically corresponds withthe results of other researches and provides effective evidence for sreearch on biochemical basis in the pollen abortion processof HPGMR.
