烟草原生质体微滴培养及细胞早期分裂的定点观察
MICRODROPLET CULTURE OF TOBACCO PROTOPLASTS AND OBSERVATIONS TRACING THEIR EARLY CELL DIVISIONS
【摘要】 <正> 原生质体培养的常规技术是群体培养。而建立单个(或少数)原生质体培养技术则可以跟踪每一原生质体的动态变化与研究不同类型原生质体之间的相互关系,从而使有关的细胞生物学研究更加精确深入;还可以有选择地单独培养细胞融合体、突变体、转化细胞等遗传操作
【Abstract】 A simple technique for culturing a fewtobacco protoplasts in microdroplets of me-dium following Koop’s method was establis-hed.Mesophyll protoplasts precultured for 2days in large population were inoculated insmall quantity (1—14 protoplasts) in 90—150 nl microdroplets.They were able to di-vide into multicellular clusters.The densityof protoplasts and the rate of early cell di-visions greatly influenced the induction resu-lts.Early development of individual proto-plasts in microdroplets was traced and descri-bed.
