脊髓灰质炎病毒中Ⅰ9株基因组克隆及其部分结构分析
Molecular Cloning and Partial of Structure Analysis of Poliovirus China I9 Genome
【摘要】 以脊髓灰质炎病毒RNA为模板,反转录合成了长链ds-cDNA。将合成的脊灰病毒中I9株ds—cDNA片段重组到质粒pAT153上,获得了约占脊灰病毒中I9基因组95%以上的cDNA克隆。对克隆的中I9cDNA部分片段作了限制性内切酶图谱分析和部分核酸的序列分析,发现脊灰病毒中I9部分核酸顺序有所改变。
【Abstract】 The optimal conditions of ds-cDNA synthesis of long chain molecule with poliovirus RNA were compared with Gublers method(5). It was found that there was remarkable influence on the sythesis rate and segment size of the second-stranded cDNA when a specific amount of RNaseH was used. Poliovirus ds-cDNA of China I9 strain was ligated to plasmid pAT 153 using EcoRI linkers, Clones were screened and the cDNA of about 95 per cent of China I9 genome was obtained.It was found that the PstI, BamHI and HindⅢ sites on the China I9 cDNA were identical with those on the type I cDNA (Lsc,2ab and Mahoney) .Partial sequence analysis of PO7 China I9 cDNA revealed that the genome structure of China I9 strain was a variant of that of other type I strains.
