白僵菌液体培养物中甾体基质和产物的薄层色谱及光密度扫描测定

【摘要】 <正> 为测定白僵菌液体培养物中基质Reichsteins化合物S(I)和它的11α-羟基化产物表皮质醇(Ⅱ),我们建立了薄层色谱(TLC)及光密度扫描测定法。现将本方法报告如下。

【Abstract】 Method of thin-layer chromatography(TLC) with subsequent deasitometric scanning has been developed for the determination of steroid substrate, Reichstein’s S(RS), and its lla-hydroxy-lated product, epicortisol, in the fungic extracts of Beauveria bassina AS69.The method requires initial extraction of the culture broth media with ethyl acetate. Extracts are submited to TLC development on silica gel GF254 layer using three solvents. The solvent systems provide a good separation of RS and epicortisol. Quantitative analysis of steroid substrate and its lla-hydroxylated product by TLC with subsequent densitometric scanning is accurate and precise. The TLC method is applied to the study of microbial hydroxylation of RS by the organism of Beauveria bassina AS69.
