化学合成的人α-心房肽基因在SUC2启动子-信号顺序控制下的表达
EXPRESSION OF CHEMICAL SYNTHESIZED α-hANP GENE DIRECTED BY SUC2 PROMOTER-SIGNAL SEQUENCE
【摘要】 本文报道将化学合成的人α-心房肽(α-hANP)基因与酵母分泌型表达载体YFD6A21重组,使α-hANP基因在蔗糖酶基因(SUC2)启动子-信号顺序指导下,在酵母菌中合成和分泌有活性的α-hANP。放射免疫分析表明:α-hANP基因只有在葡萄糖去阻遏条件下才能得到表达,而且其表达量与细胞浓度成正比。此外,95%以上表达产物被分泌到培养液中。
【Abstract】 The Chemical synthesized α-hANP gene has been inserted into yeast expression vector YFD6△21. The expression of α-hANP gene was directed by SUC2 promcter-signal sequence. The results of radioimmunoassay showed that the yeast cells containing chimeric vector can synthesize α-hANP only at glucose derepression condition, and the expression level was related to the cell concentration. In addition, more than 95% of expressed α-hANP was secreted into the medium.
