多花黑麦草原生质体培养和再生白化苗

【摘要】 本文报道从多花黑麦单(Lolium multiflorum Lam.)原生质体培养获得愈伤组织并再生白化苗的试验结果。利用多花黑麦草幼穗在MS+3mg/L2.4-D的培养基上诱导出愈伤组织,并在MS+2mg/L2.4-D的液体培养基中振荡培养,产生悬浮细胞系。从悬浮细胞制备原生质体,用看护培养,琼脂糖固体平板培养和液体浅层培养均获得了再生细胞团。这些细胞团在MS+1mg/L2.4-D+60g/L蔗糖的培养基上形成愈伤组织和胚状体,并在分化培养基上再生出白化苗。试验还对有关培养基进行了研究。

【Abstract】 Protoplasts isolated from suspension cells of annual ryegrass (Lolium multiflorum Lam.cv.Multimo) were used to investigate callus formation and plantlet differentiation. The suspension culture was initiated in MS+2mg/L 2,4-D medium from young inflorescence calli which were cultured in MS+3mg/L 2.4-D medium. The regenerated protoelones were obtained through mixed nurse, agarose plate and liquid shallow layer culture methods. The calli and embryoids were formed from the protoelones in MS +1mg/L 2.4-D+60g/L, sucrose medium. The albino-plantlets have been differentiated from the embryoids in regeneration medium. Several protoplast culture media were used and studied in the experiments.
