【摘要】 本文应用淋巴细胞毒、间接免疫荧光、流式细胞分离仪和淋巴细胞转化抑制试验测定了三株杂交瘤细胞所分泌的抗小鼠T细胞表面Thy—1、L3T4、Lyt—2单克隆抗体的特异性和效价。发现抗L3T4和抗Thy—1单克隆抗体对胸腺和脾脏细胞的识别率为95％和25％;抗L3T4和抗Lyt—2单克隆抗体对胸腺细胞的结合率分别为80％和90％;对脾脏细胞结合率分别为18％和11％。经抗Thy—1和抗L3T4单克隆抗体加补体处理后的脾细胞对ConA刺激的增殖能力明显被抑制,而对LPS刺激的增殖能力正常。本文结果除抗L3T4、Lyt—2单抗的细胞毒试验未见报导外,其它结果均和原作者的报导符合。更多还原

【Abstract】 Cytotoxicity assay, indirect immunofluorescence, fluorescence-activated cell sorter and lymphocyte transformation inhibition test were used to determine the titration and Specificity of the anti-Thy-1, anti-L3T4 and anti-Lyt-2 McAbs secreted by 20-10-5S, GK 1.5, 53-6.72 cell lines. It was found that anti-Thy-1 McAb combines with 95% of thymus cells and 25% of spleen cells; anti-L3T4 and anti-Lyt-2 bind with 80% and 90% of thymus cells, 20% and 10% of spleen cells. Anti-Thy-1 and anti-L3T4 McAbs plus complement can completely block the responce of spleen cell to ConA stimulation while the responce to LPS was normal. Anti-Lyt-2 McAb blocks the responce of the spleen cell neither to ConA nor to LPS. All of the results were conformed with previous reports except for the result of the cytotoxicity assay of the anti-L3T4 and anti-Lyt-2 McAb which were not reported.更多还原
