5-Br-PADAP-Triton X-100析相光度法连续测定生物试样中的痕量铜和锌

【摘要】 鉴于Triton X-100在高于其表现浊点下对铜和锌与[2-(5-溴-2吡啶偶氮)-二乙氨基苯酚](简称5-Br-PADAP)配合物的析相富集作用,利用铜试剂(DDTC)分解铜的配合物而褪色,锌的配合物不被分解,借此差减法实现了铜、锌的连读测定。试验表明,在掩蔽剂存在下,控制pH8.0~10.0范围内,Zn(Cu)-5-Br-PADAP配合物的λmax=560nm,εCu=1.14×105Zn=1.47×105,铜锌含量分别为0~5μg/5ml、0~4μg/5ml,服从比耳定律。方法用样少,无需特球仪器,简便、灵敏、选择性较好,适用于血清、头发、粮食中的痕量铜、锌连读测定。

【Abstract】 Traces of copper and zinc can be enriched as chelates with 5- Br-PADAP in the presence of Triton X-100 by phase separation above its apparant cloud point. The total absorptivity of 5-Br-PADAP chelates of Cu (Ⅱ) and Zn (Ⅱ) is first measured, and the absorptivity of Zn (Ⅱ)-chelate is measured after the addition of DDTC to decompose the chelate with Cu(Ⅱ). the absorptivity due to Cu(Ⅱ)-chelate can be obtained by diffcrcnce. In the presence of masking agents and with the maintenance of acidity at pH 8.0~10.0, the apparant molar absorptivity for Cu (Ⅱ)-chelate and Zn(Ⅱ)-chelate were found to be 1.14×10~5 and 1.47×10~5 correspondingly. Beer’s law is obeyed in the range of 0~5μg Cu/5ml and 0~4μg Zn/5ml. This method has been applied to the analysis of blood serum, hair and grain.
