抗hCG单克隆抗体对hCG和LH刺激Leydig细胞分泌睾酮的影响

【摘要】 本工作观察了五种抗hCG单克隆抗体对hCG/LH刺激BALB/c小鼠Leydig细胞分泌睾酮的影响。抗整个hCG分子的抗体2C5*及抗hCH-β亚单位的抗体3C6*和2A4*对hCG及LH刺激睾酮分泌均有抑制作用。抗整个hCG分子的抗体hCG1*可阻断hCG刺激睾酮分泌,而对LH则无阻断作用。抗hCG-β亚单位羧基端37肽片段的抗体3A4*既不能抑制hCG,也不能抑制LH刺激睾酮分泌。结果提示在hCG分子上存在着特异的与其生物学活性表达有关的抗原决定簇。

【Abstract】 In our Study, we employed five of monoclonal anti-hCG antibodies prepared in our laboratory. hCG1 and 2C5 are McAbs to hCG, 3C8 and 2A4 are McAbs to hCG-β, 3A4 is a McAb to C37 on hCG-β.2C5, 3C8 and 2A4 could inhibit not only hCG but LH inducing testosterone secretion in leydig cells of BALB/c mouse, hCG1 could only inhibit hCG, whereas, 3A4 could inhibit neither hCG nor LH.The results suggest that there are unique epitopes of hCG.In this experiment, the steroidogenesis was monitored by radio-immunoassay
