神经降压素对大鼠胰岛B细胞损伤的预防作用
Preventive Effect of Neurotensin on Islet B-cells Against the Damage Produced bs Streptozotocin and its Preliminary Mechanism
【摘要】 腹腔注射链佐霉素破坏大鼠胰岛B细胞以诱发糖尿病,注后24小时,血糖明显升高,血清及胰组织中胰岛素含量降低、其离体灌流的胰腺对高糖刺激的胰岛素释放反应也显著降低。10分钟前先给予神经降压素,对上述反应有明显的抑制作用,并具有量效关系。组织学检查证明,神经降压素预防注射可减轻链佐霉素对B细胞的损害。由于阿托品及酚妥拉明均可部分阻断神经降压素的这一作用,提示胆硷能M受体和肾上腺素能α受体可能参与神经降压素对B细胞的保护作用。
【Abstract】 Twenty-four hours affer injection of streptozotocin (45mg/kg) (NS + STZ) in rats showed a significant increase in serum glucose and a great decrease in serum and pancreatic insulin. These changes were not observed in animals pretrcated with neurotensin (40μg/kg, S. C. ) (NT + STZ) . Neurotensin injected alone exhibited no significant effect on serum glucose concentration. Statistical differences of serum glucose, serum insulin and pancretic insulin between NS+STZ group and NT + STZ group were significant. In the perfused pancreas isolated from rats treated with NS + STZ,high concentration of glucose (19.7m.M/L) could not induce increased secretion of insulin to the perfusate, but a sharp insulin secretory peak was found in preparation treated with neurotensin and STZ. The morphological changes of the pancreatic islets were also examined. The insulin secretory granules in B-cells were decreased obviously in the NS+STZ group, while in the NT+ STZ group they were almost normal. Thus it is apparent that neurotensin can protect the function of islet B-cells against the damage produced by streptozotocin. In addition, the antagonists as atropine and phentolamine could partially block the preventive action of neurotensin, suggesting that the muscarinic and α-adrenergic receptors might take part in the preventive action of neurotensin. neurotensin streptozotoc in pancreatic islet B-cells diabets insulin
