用幼穗组织培养筛选小麦耐盐突变体
THE SELECTION OF NaCI-TOLERANT MUTANTS BY TISSUE CULTURE IN WHEAT
【摘要】 运用组织培养研究了小麦耐盐(NaCl)突变体的筛选,获得了大量耐盐细胞系,其中有5个系的耐盐性稳定而且较强,其余的系在以后继代培养中耐盐性逐步消失。耐盐稳定的细胞系在含NaCl的培养基中再生了植株,其中三棵植株表现了耐盐特性,而且两株的耐盐性通过有性世代仍保留,初步确定为遗传的耐盐特性突变体。在试验中采用了60Co γ射线辐照幼穗,结果表明1kR比较有效。
【Abstract】 The selection of NaCl-tolerant mutants in wheat was studied,A number of NaCl-tolerant cell lines were selected from a great quantity ofcallus cultures.The NaCl-tolerance of 5 lines were stable and very strongafter subculture of three times. The plants were regenerated on 1.0%NaCl-containing media.The NaCl-tolerance appeared in the callus from embryos of three plants regenerated.Two of the three plants which could pass on the NaCl-tolerance to next generation through gametophyte are preliminarily considered as NaCl-tolerant genetic mutant. 60Co γ-ray was used to induce the mutation and 1kR was found to be effective.
