微载体培养Vero细胞工艺条件初探

【摘要】 本文研究了细胞生长过程中微载体的种类,浓度及细胞的接种量、溶解氧水平对培养Vero细胞的影响。在pH为7.0~7.4,液解氧水平为30%~50%空气饱和浓度,搅拌速度为25~35r/min,温度为37℃时,在1.5升CelligenTM系统中,连续培养7天,可使细胞浓度达到2.5×106 Cells/ml,为接种量的12倍,细胞生长旺盛,形态正常。

【Abstract】 Several factors which may affect the growth of Vero cell have been studied. The kind and concentration of microcarriers, cell inoculation density and dissolved oxygen level can influence cell growth substantially. Under the following conditions: pH=7.0~7.4; dissolved oxygen, 30%~50% saturated air; stirring speed, 25-35r/min; and temperature 37℃, cell concentration can reach 2.5×10~6 cells/ml after 7 days culture in the 1.5 L CELLIGEN system. The resulting cell concentration is almost 12 times as high as the initial cell inoculation density.
