赤霉素产生菌~#218菌株的研究——Ⅰ.原生质体的分离和再生

【摘要】 赤霉素产生菌~#218菌株菌丝体用玻璃纸平板培养法培养,培养时间为36小时。采用2%蜗牛酶,不须硫醇类化合物预处理,可直接裂解赤霉素产生菌~#218菌株菌丝体,得到大量的原生质体。将原生质体悬浮液接种到高渗性再生培养基上,再生频率约为1~2%。原生质体再生菌株仍保留亲株的菌落形态及合成赤霉素的生产能力。

【Abstract】 Fusarium moniforme ~#218 strain protoplasts were obtained from cultures grown on a agar plate at 28℃ for 36 hours. Their isolation and regeneration have been investigated. Protoplasts can be isolated from F. moniforme more readily after treatment of myclium with 2% snail digestive juice (helicase) and without treatment than after treatment with DTT. Regeneration frequency of purified protoplast suspension plated on a highly osmotic RM is about 1~2%. The yield of Gibberellin and morphology of colonies of the regenerated strains from protoplasts are the same as those of the parent strain.
