【摘要】 本研究创立一种灵敏、准确的甲胎蛋白定量方法酶联火箭电泳测定法。此法是将辣根过氧化物酶标记的抗甲胎蛋白抗体混合于琼脂糖凝胶中进行火箭电泳,免疫沉淀线具有酶活性,经与 H₂O₂和3-氨基-9-乙基卡巴唑保温,借酶促反应使沉淀线出现强烈的有色沉淀而被清楚显示出来。这种新方法可对甲胎蛋白作出灵敏、快速而精确的定量。本法甲胎蛋白定量的下限为5～10ng/ml,灵敏度比火箭电泳法高100倍,比放射火箭电泳法高2～5倍。其甲胎蛋白测定范围为10～3000ng/ml,宽于放射火箭电泳法（25～800ng/ml）和放射免疫测定法（RIA）（0～400ng/ml）。本法准确度与RIA 一致。组内变异系数为2.4%,组间变异系数为3.1%,而 RIA 法为17.4%。更多还原

【Abstract】 A sensitive and accurate quantitative method of AFP,enzymorocket electrophoresis,was developed by introducing horseradish peroxidase-labeled anti-human AFP antibody into rocket electrophoresis.The immunopre- cipitate,which has been enzymatically active,is visualized by incubation with H₂O₂ and 3-amino-9-ethylearbazole,which stains the immunopreci- pitate with a strongly coloured deposit.This new method gives sensitive, rapid and precise quantification of AFP. The lower limit of quantitation by this method is about 5～10 ng/ml of AFP.Compared with the rocket eleetrophoresis and radioroeket eletropho- resis,there are respectively approximately 100-fold and 2-to 5-fold increase in sensitivity.The dose-response curve covers a broader range of concentrations of AFP（10～3,000ng/ml）than RIA（0～400ng/ml）and radioroeket eleetrophoresis（25～800ng/ml）.The accuracy of this method is almost comparable to that of RIA.The within-run coefficient of varia- tion is 2.4% and the between-run coefficient of variation is 3.1%.更多还原
