棉籽饼中残留棉酚的反相高效液相色谱分析
DETERMINATION OF RESIDUAL GOSSYPOL IN COTTON SEED CAKE BY REVERSED-PHASE HPLC
【摘要】 本文介绍一种测定棉籽饼中残留游离棉酚的反相高效液相色谱法并和紫外分光光度法相比较。用四氢呋喃提取棉籽饼中游离棉酚,然后在在μ-Bondapak C18柱上进行分离,采用甲醇-水-磷酸(82:18:0.1)作为流动相。样品制备方法简单,回收率高,最小可检测量约为10ng,可测定棉籽饼中低至1μg/g的游离棉酚。
【Abstract】 A reversed-phase HPLC method for the determination of residual gossypol in cotton seed cake was developed and compared with the current spectrophotometric method. The free gossypol was extracted with tetrahydrofuran, and then separated on μ-Bondapak C18 column with methanol-water-phosphoric acid(82:18:0.1)as the eluant. A good linear calibration curve was obtained by using either peak height or peak area over the experimental concentration range from 6.87μg/ml to 96.1 μg/ml with the minimum detectable limit of ca. 10ng.The procedure of sample preparation was simple and the recovery was high. The results showed that the method could be used for the determination of as low as 1 μg/g gossypol in cotton seed cake.
