咖啡因分别与137Csγ射线和氚水复合作用对CHL-1细胞的增殖和恶性转化的影响
EFFECTS OF THE COMBINATIONS OF CAFFEINE WITH 137Cs-GAMMA RAYS OR TRITIATED WATER ON THE PROLIFERATION AND MALIGNANT TRANSFORMATION IN CHL-1 CELLS
【摘要】 本实验观察了咖啡因分别与137Csγ射线和氚水复合作用对 CHL-1细胞的增殖抑制和恶性转化效应的影响。在咖啡因浓度为1—2mmol/L,137Csγ辐射剂量为0.837—2.51Gy,氚β剂量为0.088—0.528Gy 条件下,上述两种射线或咖啡因各自单独作用,以及咖啡因分别与上述射线复合作用时,均可使 CHL-1细胞增殖受抑制,抑制的程度与辐射剂量和咖啡因的浓度有关。转化实验中各实验组的细胞恶性转化率都比对照组高,咖啡因分别与上述两种射线复合作用的实验组,其转化率又比各自单独作用时高。把转化了的细胞注入经照射后的小鼠,部分动物出现约2mm3大小的肿块,肿块结构的细胞形态与体外培养的转化了的细胞形态相同
【Abstract】 The effects of the combinations of caffeine with 137Cs-gammarays or tritiated water on the proliferation and malignant transformationin vitro in CHL-1 ceils were observed in experiments.At the concentrationsof caffeine from i mmol/L to 2 retool/L,the dose ranges of137Cs-gammarays from 0.837Gy an d to 2.51 Gy and of tritium-beta radiationfrom0.088Gy to 0.528Gy,the cell proliferation of CHL-1 cells was found to beinhibited when cells were exposed to caffeine,gamma and beta radiations,respectively,as well as when they were exposed to various combinations ofcaffeine with the two latters.The degree of inhibition of cell proliferation-was dependent upon the concentration of caff eine and on the doses of-radiation.In the transformation experiments,cell malignant transformationrates for all treated groups were higher than that for contol group and therates for irradiated plus caffeine-treatedgroups were higher than those for-corresponding single-agent-treated ones.After the subcutaneous injectionof transformed cells into irradiated mice,tomours in size of about 2mm3 were found in some animals and the tomout cells werei denticalwith in-vitro-transformed CHL-1 cells histopathologically.
