6—BA、IBA激素对花叶芋(Caladium bicolor)外植体愈伤组织的诱导及器官分化的影响

【摘要】 在花叶芋的组织培养中,以生长健壮的叶片、叶柄为外植体、在附加6—BA1ppm;6—BA2ppm+IBA0.5ppm;6—BA1ppm+IBA2ppm的MS培养基上,均能诱导出愈伤组织;其中以6—BA2ppm+IBA0.5ppm的诱导率最高。试验发现,使用低浓度的IBA加高浓度6—BA的MS培养基,愈伤组织诱导率及后期培养的再分化率都较高。愈伤组织转移到附加6—BA3ppm的MS培养基上分化率达105%。分化培养中愈伤组织也在不断增长,并同时在芽的基部长出根系,形成完整植株,使整个培养周期缩短一个月。用无土栽培法培养过渡幼苗,生长极快,能在较短的时间内达到具有商品性能的健壮植株。

【Abstract】 Health foliage and petiole as explants on the MS medium supplemented 6—BA (lppm), 6—BA(2ppm)+IBA(0.Sppm), 6—BA(1ppm)+IBA (2ppm) could all produce callus, and the highest one of inducing frequency was 6—BA (2ppm)+IBA(0.5ppm) among above three hormone combinations. The experimental results showed that inducing frequency of callus and redifferentiation frequency at later period were all higher if medium of low IBA conc. plus high 6—BA conc. were used. The frequency of differentiation was 105% when the callus was transported onto MS medium supplemented 6—BA (3ppm). The callus was increasing all the time in differentiation culture, and many roots were produced on the base of shoots. Whole plantlets were formed and the period of culture was shorten into a month. The transition seedlings grew quickly while using soilless culture method and became health plants with commercial characteristics within a short time.
