苯妥英钠对发育期小鼠小脑形态和体格发育的影响

【摘要】 生后4天龄正常幼鼠持续应用苯妥英钠50天,观察该药对发育中的小脑形态及体格发育的影响。25mg/kg和50mg/kg两个剂量组动物的小脑,在光镜下均未见明显病理改变,小脑皮层浦肯野氏细胞计数各为141.7±1.99/20um~2和13.62±1.93/20um~2,与对照组比较均无统计学意义。各组动物的体重无明显差异。说明苯妥英钠在本剂量范围及给药时程内对发育期小鼠的小脑形态无明显损害作用,也不影响其体格发育。

【Abstract】 We treated black mice pups with diphenylhydantoin (DPH) continually for 50 days. The chronic effects of the drug on the morphology of developing cerebellum and physical development were investigated. The mice treated at 25 mg/kg/dose q. d. and the mice treated at 50 mg/kg/dose q. d. had serum drug levels of 12.7 ug/ml and 24.8 ug/ml, respectively. No cerebellar pathological changes were found in either of the groups of animals under light microscope. The numbers of Purkinjc cells in both the low dose group and the high dose group failed to show any statistical differences as compared with the control (P>0.5). There were no differences in physical development as shown in body weights of the treated and control mice at 54 days of age. The results suggested that DPH given continually during a period of 50 days within the 25-50 mg/kg dosage range had no significant effects on cerebellar morphology and physical development in suckling mice.
