三种抗卵巢上皮癌单克隆抗体的制备及其性能的研究与比较

【摘要】 用卵巢上皮癌可溶性抗原,制备了三株分泌抗卵巢上皮癌单克隆抗体杂交瘤细胞株,分泌抗体稳定。对其抗体亚类、效价及亲和力进行了测定,经免疫组化检测对三种抗体进行了比较,并碘标一株单克隆抗体进行荷瘤裸鼠放射免疫显像,结果表明特异性良好,为进一步应用展示了良好前景。

【Abstract】 Three murine monoclonal antibodies COC 166-9, COC 183-B2 and COC 183-A5 were raised against human epithelial ovarian adenocarcinoma associated antigens by fusion of murine myeloma cells with spleen lymphocytes from BALB/C mice immunized with soluble antigens prepared from two ovarian papillary serous cyst adenocarcinomas. The monoclonal antibodies were stable after culture and storage under 80× for 20 and 11 months. The subclass were all IgG1. The assays of the liter and affinity of the 3 monoclonal antibodies were 1 × 105, 2×l05, .8×105 and COC 166-9>COC 183-B2>COC 183-A5. me reactivities were tested on varous paraffin embedded tissue sections by immunoperoxidose stainings, whereas the ratios of postive staining with ovarian epithelial adenocarcinoma were 77.8% for COC 166-9, 87.2% COC 183-A, and 75.6% COC 183-B2 respectively.Human ovarian carcinoma xenograft in nude mice wero radioimaged 48 hours after 131I-COC 183-B2 administration, gamm,a scimigraphy demonstrated specific localization of the radiolabeled monoclonal antibody in these tumors. The above results showed a bright future for immunodiagnosis by means of these monoclonal antibodies, as well as immunotherapy.
