小鼠胚泡着床过程中子宫6-酮-PGF1α(6-KF)及TXB2含量的变化

【摘要】 本实验利用前列环素(Prostacyclin,简称PGI2)及血栓素(Thromboxane A2,简称TXA2)的代谢产物6-KF及TXB2的RIA技术,探讨了小鼠子宫在胚泡着床点及非着床部位上述二类PGs的含量变化。实验表明,妊娠D5胚泡着床部位及着床间区,6-KF的水平较高;而TXB2含量较低变化也小。表明胚泡着床时,血管通透性增强与PGI2升高密切相关。PGI2与TXA2比值的增高,使血管舒张,增强抗凝,有利于胚泡着床及营养供应。

【Abstract】 The concentration of 6-KF and TXBa in the implantation site and non-implantation region of the mouse uterine tissue were measured by RIA. For comparison, pregnant and pseudopregnant animals were used. In pseudopregnancy, the uterine horns of mice were traumatized with silk thread to induce deciduoma in comparison with the normal implantation site. Animals were killed on D5 and Ds after coitus. It was shown that, the non-implanted region of pregnant uterus contains a large quantities of 6-KF. There is a significant increase (P<0.05) in the pregnant uterine content of 6-KF during implantation than the same time of pseudopregnan-cy. The levels of 6-KF reach a peak at non-implanted region (149.66+++++++++++++++++24.06pg/mg) and in higher level at implantation site (95.76+++++++++++16.80pg/mg) respectively on D5 and decreased on D6 (52.36+++++++++++++++6.12 and 45.45 + 8.46pg/mg). TXB2 concentration showed only small changes during implantation, but in the tissue of traumatized, pseudopregnant mice, TXB2 contents change from a lower level in D5 (24.74++++++++++++9.73, 37.72++++++++++++12.94pg/mg) to a higher values in D6 (68.56+++++++++++12.15, 89.06+++++++++++++9.94pg/mg). No significant changes of TXB2 were observed in uterine tissue of pregnant mice (P>0.05). Presumably the elevated levels of 6-KF reflect increased localised synthesis of PGI2 in the myometrium. It may be involved in the mediation of increased vascular permeability, which acts an important role in the process of implantation. After the nidation of blastocy-stes, the levels of 6-KF decreased. The mechanism of PGI2 synthesis has been discussed.
