【Abstract】 To map the polyhedrin gene of the PlNPV, AcNPV -DNA EcoR I -I fragment was used as a probe to hybridize with the P1NPV-DNA restriction fragments immobolized on nitrocellulose filte under stringent and nonstringent conditions. Under stringent condition (68℃),no hybridization was found between the probe and the P1NPV-DNA. But under nonstringent Condition (35℃, 20% formamide), the P1NPV-DNA EcORI-6.8kb fragment can hybridize with the probe. By using a range of formamide. concentration to vary the hybridization conditions, the Tm value (71℃), mismatch rate(l8%),and polyhedrin gene homology(82%)of the P1NPV-AcNPV DNA hybrid was evaluated.更多还原
