【摘要】 同时应用大剂量醛氢叶酸(CF)可增加氟嘧啶(5Fu)的抗肿瘤作用。我们以CF 200毫克/日合并5Fu365-500毫克/平方米/日静脉注射,每日一次连用五日为一疗程。共治20例病理证实为晚期胃肠道癌病人,13例曾单用5Fu或FT-207治疗无效。19例可评价疗效5例胃癌治疗后有1例部分缓解,1例微效,3例稳定。14例结直肠癌治疗后3例部分缓解,5例微效,4例稳定,2例恶化。本疗法的主要毒性包括恶心呕吐、口腔炎、食欲不振、腹泻、白细胞下降和脱发等。这些副作用似乎比单用5 Fh时为重。本探索性研究可见,HDCF/5Fu疗法是晚期胃肠道肿瘤的有效治疗方法,但应仔细注意毒副作用的发生。更多还原

【Abstract】 20 cases of histologically confirmed advanced G-I cancer patients, all prev iously treated with 5-FU, were treated with a combination of high dose iv citrovorum factor (200 mg/d) and 5-FU (365-500 mg/M2/d) for 5 consecutive days. 19 cases were evaluable^or response. Of 5 gastric cancer cases, 1 achieved PR, 1 MR and 3 SD. Of 14 colorectal cancer cases, 3 achieved PR, 5 MR, 4 SD and 2 PD. Main toxic side effects included nauseavomiting (9),. stomatitis (9), anorexia (13), diarrhea (5), leucopenia (10) and alopecia (8). which were considered to be more severe than what usually seen with 5-FU used alone. This pilot study showed that HDCF/5-FU is an effective treatment for advanced G-I cancer, however, caution should be taken for the exaggerated side effects.更多还原
