缩二脲法分光光度测定豆类和谷类蛋白质

【摘要】 本文提出了一种测定豆类和谷类粗蛋白质的简单、快速方法—缩二脲法。该法使用0.1NNaOH溶液溶解样品,以大牛血清白蛋白作为标准蛋白质,利用缩二脲试剂分光光度测定样品中的蛋白质含量。紫红色配合物的最大吸收波长为555nm。比色液中蛋白质含量为0~3mg/ml时,符合比尔定律。测定结果与凯氏法非常接近,其精密度和准确度也是令人满意的。

【Abstract】 A simple and rapid method for determining of the crude proteins in the beans and cereals, biuret method, is presented in the paper.With the sample dissolved in 0.1NNaOH solution and bovine serum albumin used as the standard protein, the Percentage of proteins in the sample is given by spectrophotometric method with biuret reagent.The maximum absorption wavelength of the violet red complex is 555nm. If the relationship between the concentration of proteins of the color solution and the absorbance, is in the concentration region of O-3 mg/ml of proteins, it obeys Beer’s law. The determinated results are very conformable to the values obtained by Kieldahl method.Its precision and accuracy is yet satisfactory.
