利福喷丁侧链N-环戊基-N′-氨基哌嗪的气相色谱分析
GAS CHROMATOGRAPHY ANALYSIS OF THE SIDE CHAIN OF RIFAPETIN
【摘要】 长效抗结核抗生素利福喷丁侧链-N-环戊基-N′-氨基哌嗪的气相色谱分析的结果表明,该侧链和其它13个哌嗪类化合物,使用2%OV-17-chromosorb W作固定相能获得完全分离.由于侧链不够稳定,采用苯甲醛同侧链氨基缩会为N-环戊基-N′-苄亚氨基哌嗪的衍生化方法能够准确、快速、简便地定量分析侧链.该衍生化方法分析侧链比其它衍生化方法优越并能获得满意的定量结果.
【Abstract】 This paper dealt with the gas chromatographic analysis of N-cyclop-entyl-N’-amino-piperazine——the side chain of long-acting antituberculosis antibioticRifapentin.Experimental results showed that the side chain and other 13 related piperazine compounds in the sample could be completely resolved by using 2% OV-17 on chromosorb W (60-80 mesh)as stationary phase.A derivatization method was proposed due to the labile property of the side chain, by condensing the amino group of the side chain with benzaldehyde to form Ncyclopentyl-N’-benzalimide-piperazine.This method is accurate, rapid and simple. It can be used for quantitative analysis of the side chain and is superior to silyation and acetylation method. The derivatization procedure reacted quantitatively, and the product is a stable solid compound which is easily purified.The quantitative standard deviation is 0.6 and relative standard deviation is 0.7%.
