头孢三嗪与妥布拉霉素协同抗菌作用机理研究 Ⅰ.头孢三嗪与妥布拉霉素对细菌胞壁及蛋白质合成的影响

【摘要】 头孢三嗪能迅速抑制[3H]-DAP掺入大肠杆菌K12 DP50 Sup F,阻碍DAP参与该菌的胞壁形成与细菌生长.头孢三嗪与妥布拉霉素有抗菌协同作用.但头孢三嗪对[3H]-亮氨酸掺入绿脓杆菌10118无明显抑制作用.妥布拉霉素对[3H]-亮氨酸掺入绿脓杆菌呈明显抑制作用,抑制曲线随药物浓度不同而呈三相变化.头孢三嗪与妥布拉霉素合用[3H]-亮氨酸掺入绿脓杆菌10118的作用比单用妥布拉霉素弱,但曲线亦呈三相变化.结果表明;头孢三嗪作用机理在于抑制胞壁前体物质粘肽的合成,而对蛋白质合成无明显影响;而头孢三嗪与妥布拉霉素协同作用机理可能是诱导或促进了细菌蛋白质的合成,而并非是增强了对细菌蛋白质合成的抑制作用.
【Abstract】 The ceftriaxone and tobramycin on the synthesises of cell wall and protein were reported. In E.coli K12 DP50 Sup F, incorporation of [3H] diaminopimelic acid (DAP) was strongly inhibited by ceftriaxone, but incorporation of [3H] leucine in P.aeruginosa 10118 was slightly inhibited.From these results,it was inferred that the ceftriaxone primarily inhibited cell wall synthesis. Tobramyctn inhibited protein synthesis in the triphasic way. Ceftriaxone and tobramycin in combination inhibited protein synthesis in the way similar to tobramycin type, but slightly less than tobramycin acted singty. It may be referred that the synergism of the two drugs is not by inhibitiou of the protein synthesis.
