青霉素酰化酶的百里酚红测定法—NIPAB改良法
THE THYMOL RED METHOD FOR DETERMINNING PENICILLIN ACYLASE ACTIVITY桝N IMPROVEMENTOF THE NIPAB METHOD
【摘要】 本文介绍我们建立的一种青霉素酰化酶测定方法,是对NIPAB法的一种改进。其反应特异性高,操作简便,检测灵敏度较原法高约三倍。既可用于青霉素酰化酶的分光光度法测定,也能用于产酰化酶菌株的平板筛选。百里酚红于 510nm的克分子消光系数为2.8× 104l·mole-1·cm-1。
【Abstract】 In this paper a new method for determination of penicillin acylase (E. C 3. 5. l. 11)activity is proposed. It is an improvement of the NIPAB method having advantages of high specificity, convenient in operation and 3-fold in creaci-ng in detection sensitivity. The method can be applied either in solution as a conventional spectrophotometric assay or on plate for selection of those penicillin acylase producing bacteria. The molar absorbance of the thymol red determined is 2.8×10~4 l.mole~-1 ·cm~-1.
