免疫胶体金试剂的制备及淋巴细胞膜抗原的检测

【摘要】 本文报道采用国产试剂,将胶体金标记在葡萄球菌蛋白A(SPA)和纯化免抗鼠IgG抗体(RAM)上,制备了数批免疫胶体金探针试刺,并建立了免疫金银染色法,用于人T、B细胞膜表面抗原的光镜水平检测。阳性细胞膜上出现的免疫金银颗粒有三种类型:均匀分布的细小颗粒、局部粗大颗粒、以及帽状聚集的颗粒。次外,T、B细胞膜抗原的检出阳性率与以往报道的免疫荧光法相符合。因此,免疫金银法是一种敏感、可靠、方便和值得推广的方法,为研究细胞表面标记提供了一个新的技术途径。

【Abstract】 This paper introduces a method to label protein A and rabbit antimouse IgG on colloidal gold by using some reagents made in China and an immunogold silver staining procedure to identify antigens on human lymphocytes in optical microscopy. The deposited particles of immunogold silver compound menifests themselves as numerous small scattered black dots, localized large black dots, and cap-like aggregations on the positive T and B cells. The positive rate detected by this method is in agreement with that by immunofluorescence technique, and this method is thus proved to be sensitive, rapid and reliable, worthy to be popularized. Therefore, a new technical approach to the detection of antigens on cell surface membrane is now available.
