人脑多形性胶质母细胞瘤细胞系“BT325”的建立及其生物学特性

【摘要】 本文报道一例人脑多形性胶质母细胞瘤细胞系“BT325”的建立及其生物学特性,至今已传180代,维持4年以上,生长稳定,每周传代一次,细胞形态多样,星球状,双极形,星形及少量多核巨细胞。透射电镜可见多量微管及束状微丝。异种移植至裸鼠皮下生长肿瘤,病理切片经 HE、PTAH 及 Foot’s 染色均证实为恶性胶质瘤。免疫化学染色胶质纤维酸性蛋白(GFAP)呈阳性反应,生长曲线计算倍增时间为36.5小时,染色体组型显示亚四倍体,90%以上显示有标记染色体。细胞周期时相分析用FCM 法计算出各时相的百分比为 G1=52.94%,S=28.08%,G2+M=18.98%,用 PLM 法计算出各时相的时间为 Tc=37.5小时、TG1=16.2小时,Ts=10.5小时,TG2+M=10.8小时,TM 估计值为0.8小时,细胞冻融存活率为77%左右,认为本系细胞生长和特性稳定,可作筛选化疗药物,有一定帮助。

【Abstract】 A human glioblastoma multiforme cell linehas been established from a glioma on thefrontal lobe of a senior man.This cell line wasdesignated BT 325 and has been maintained inculture for over four years.The morphologicfeatures of cells are variable.Globose,bipolar,stellate and multinuclear giant cells have beenobserved from the culture.Doubling time cal-culated from the growth curve was approximate-ly 36.5 hours.The karyotype was shown hypo-tetraploid and had one marker chromosome inmost cells.Cell cycle phase and relative DNAcontent in cells were measured from 9th to 57 thpassage by FCM and PLM.Cells inoculatedinto nude mice subcutaneously grew into tumorssimilar to the original tumour histologically.Microtubules and bundles of filaments canbe observed by electron microscope.Cells arepositively stained by antiserum to GFAP.Resultsof PTAH stain and Foot’s stain proved thesecells were malignant glioma in no time.Therate of vitality of those frozen cells around 77%showed the growth and character of the cellline was stable.It was helpful in selection ofantiglioma chemotherapy.
