免疫核糖核酸基因文库构造——免疫核糖核酸基因cDNA合成
CONSTRUCTION OF IMMUNE RNA GENE LIBRARY: SYNTHESIS OF IMMUNE RNA GENE cDNA
【摘要】 从大肠杆菌免疫的BALB/C鼠的脾脏中提取iRNA,经oligod(T)纤维素柱用亲合层析法分离poly A~+RNA,以此poly A~+RNA为模板合成了免疫核糖核酸基因的互补DNA(cDNA)。经碱性琼脂糖凝胶电泳和放射自显影分析表明,所获得的cDNA双链其分子量范围2.0~0.5kb,适合于用载体质粒构造基因文库。本文对逆转录反应和双链cDNA合成反应中的技术条件进行了讨论。
【Abstract】 In order to constract iRNA gene library cDNA was synthesized. The iRNA was extracted from BALB/C mice immunized with E. coli, and through a oligo d(T) chromotography the poly A~+ RNA was separated as the template for the cDNA. After alkalin agarose gel electrophoresis and autoradiographic analysis it was shown that the synthesized cDNA had molecular weight of 0.5~2.0kb, which would be suitable for the cDNA library.
