双向琼脂糖凝胶电泳鉴别鼠疫菌质粒构型方法的探讨

【摘要】 <正> 质粒 DNA 在制备过程中由于受损伤会出现三种不同的构型,即 CCC(超盘旋)、OC(开环)和 L(线型)。这给用琼脂糖凝胶电泳鉴定质粒带来了困难。尤其是一种细菌含有几种质粒时,在电泳凝胶中会出现复杂的DNA 区带图形。Gonzales 和 Hinterm-annts 等先后建立了双向琼脂糖凝胶电泳鉴
【Abstract】 It is simple and reliable method to apply two steps of agarose gel electrophoresis in distinction of molecular forms of Y.pestis plasmid DNA.The experimentalcount is the dose of UV illumination after the first electrophoresis.whenthe electrophoretic gel is placed at 10cm from ultraviolet lamp,it needs 10minutes’irradiate for 6 Mdal plasmid,and for 45-65 Mdal plasmid,six minu-tes is enough.But for 92 Mdal plasmid,the illuminate time is less than two min.
