【摘要】 本文报道以苜蓿银纹夜蛾核型多角体病毒多角体蛋白基因mRNA的cDNA的重组质粒PMA-Ⅵ DNA转化E.coli RR₁。采用了多种筛选方法,包括抗菌素抗性筛选,菌落杂交及电泳等方法。快速地筛选出含有PMA-Ⅵ质粒的菌株。并以蓖麻蚕NPV-DNA作探针,通过Southern杂交,表明蓖麻蚕NPV基因组中具有与苜蓿银纹夜蛾NPV多角体蛋白基因同源性的核苷酸序列。更多还原

【Abstract】 The recombinant DNA （PMA-VI DNA） containing cDNA of mRNA of polyhedringene of Autographa californica nuclear polyhedrosis virus（AcNPV） was used to transform E.coli RR₁, and colonies were selected by antibiotic resistance and colony hybridization.The colony containing PMA-VI plasmid grew in L.Broth.The plasmid DNA was rapidly isolated by boiling method, and tben recovered by electroelution. With the plasmid DNA as a probe and by Southern hybridization, the results showed that the sequence homologues to the polyhedrin gene of AcNPV was contained in the genome of Attacus ricini NPV.更多还原
