左旋四氢巴马汀对5-羟色胺所致血管收缩作用的影响
Effect of L-THP on contraction of blood vessels induced by 5-hydroxytryptamine
【摘要】 在毁脊髓大鼠,5-羟色胺iv引起舒张压升高。左旋四氢巴马汀预先ia对此升压效应具有剂量相关的明显抑制作用,其ID50为27mg/kg。以兔离休肺动脉环标本测定5-HT,NE,KCl引起的收缩反应。左旋四氢巴马汀使5-HT量效曲线平行右移,PA2值为4.64。对NE,KCl所致收缩也有拮抗作用。提示左旋四氢巴马汀具有一定的抗5-HT作用,但专一性较低。
【Abstract】 In pithed rats,iv administration of 5-HT (100μg/kg)elicited the increase in diastolic pressure.L-THP(3~100 mg/kg.ia)dose dependently reduced the increase in diastolic pressure,and its ID50 value was 27 mg/kg and cyproheptadine was 65μg/kg.The increase in diastolic pressure evoked by NE was inhibited by L-THP also,and its ID50 value was 44 mg/kg.The antagonistic effects of L-THP to 5-HT (0.3~300μM),NE(0.01~100μM).and KCl (60 mM)were studied with the isolated rabbit pulmonary artery rings.Cyproheptadine,verapamil and phentolamine were sed as reference.L-THP shifted the cumμ-ulative concentration response curves for5-HT to right,and its pA2 value was 4.64.L-THP depressed the contraction induced by NE and KCl also.As to the anti-5-hy-droxytryptamine action,cyproheptadine was stronger than L-THP.As the anti-KCl action veraparnil was stronger.As for the anti-NE action,phentoamine was more specific.It is sugested that L-THP possesses anti-5-hydroxytryptamine action on thevessels,but its specificity is relatively low.
