人红细胞的Ca2+和Cd2+运输

【摘要】 用人红细胞膜、翻膜小囊和完整红细胞研究了Cd2+对Cd2+运输的影响与Cd2+-ATP酶活性的关系,以及细胞对Cd2+的摄取和影响摄取的因素。Cd2+抑制Cd2+运输,与抑制Cd2+-ATP酶具有相关性。Cd2+极易进入细胞,一旦进入,难于排出。进入细胞的Cd2+主要与胞浆和膜蛋白相结合。

【Abstract】 In this study, we investigated the effects of Cd2+ on the Ca2 + -ATPase of human erythrocyte membranes, the Ca2 + transport in inside-out vesicles, and the Cd2 + transport in intact erythrocytes. Cd2+ inhibited the Ca2 + -ATPase activity of human red blood cell membranes, probably through an effect on the catalytic site of the enzyme. The inhibition by Cd2+ could be reversed by thiols, such as dithiothretol and glutathionine. Cd2 + had an inhibitory effect on the Ca2 + uptake by inside-out membrane vesicles.This inhibitory effect correlated well with that on the Ca2 + -ATPase. In contrast with the marked impermeability to Ca2 + , the red cells were able to accumulate Cd2 + , which was accelerated by the calcium ionophore A 23187 and glucose. As most Cd ions entering the red cells were associated with cytosol and membrane bound proteins, the cations were released very slowly into the medium.
