TritonX-100、DOC和KCI对肿瘤细胞膜抗原的增溶效果研究
Study on Solubility-enhancing Effect of Triton X-100, DOC & KCI on Tumor Cell Membrane Protein Antigens
【摘要】 肿瘤组织在—60℃连续冻融两次后,用Potter匀浆器匀浆粉碎,进行Ficoll不连续梯度离心,得纯度较高的细胞膜。用1%脱氧胆酸钠(DOC)离子去污剂、40%TritonX—100非离子去污剂和3M KCI分别进行膜蛋白质的增溶提取,以Lowry法测定提取液中的蛋白质含量。DOC提取液中蛋白质含量最高,TritonX—100次之,3M KCI最低。
【Abstract】 Tumor tissue was frozen twice at -60℃ and homogenized by Potter homogenizer. Cell membrane in homogenate was layered and purified by discontinuous Ficoll-gradient centrifugation.The cell membrane protein was extracted with 1% sodium desoxycholate (DOC) ionic detergent, and 4% Triton X-100 & 3M KCI nonionic detergents. The protein content of extracts, measured by Lowry method, extracted with DOC, was higher than those extracted with Triton X-100 and 3M KCI.
